Tissue Processing Overview

  1. 1. fixation: place in cassette, fix in formalin
    2. tissue processing (tissue is embedded in paraffin to prep for sectioning)
    -dehydration: run in automatic tissue processor overnight in series of baths of increasing concentrations of alcohol
    -clearing: remove dehydrant with xylene bath
    -replace xylene with paraffin
    -embedding: fill mold with paraffin, place tissue on mold, cool mold to form untrimmed tissue block
    3. sectioning: use microtome (tomorrow)
    4. staining (tomorrow)
